6. BRIEF RESUME OF THE INTENDED WORK:
6.1NEED FOR STUDY:
Pesticides are the compounds that are used to kill pests which may be insects, rodents, fungi, ticks etc.1

India being the second largest producer of fruits and vegetables in the world, farmers being ignorant, use harmful pesticides like Endosulfan, Aldrin, Lindane, Malathion.2


Endosulfan is known to be one of the highly toxic agricultural pesticides commonly used in our societies. With the widespread use of Endosulfan in agriculture, human beings are most likely to be exposed to it, either orally by eating Endosulfan contaminated food and whole body inhalation in the farms during its applications.3
Population that are usually susceptible to Endosulfan include the unborn, neonates, adults and the elderly people .4
The interaction of the ROS and other reactive metabolites, generated from Endosulfan metabolism, with the renal tissue may as well cause cellular injury. Hence, damage to the tissue. Once of the renal tissues are damaged, the overall functionality of the kidneys is compromised.5
Cytoprotection means protection of cell from noxious chemicals or other stimuli or Enhancing the ability of cell to resist injury.6

Cytoprotective agents will reduce or prevent these toxicities, the agents should ideally be selective for normal cell these are effective in reducing or preventing toxicity should have no negative impact.7



As per the classical text, BruhatElaishavingVishagna,Bastivikarahara, Mutrakrichrahara and Mutraghatahara properties .8
So this is an attempt made to evaluate whether BruhathEla has the cytoprotective action in nephrotoxicity and as a supportive treatment in pesticide toxicity.
REVIEW OF LITRETURE
Endosulfan is one of the synthetic organochlorine compounds widely used as agricultural insecticides. The chemical is sometimes found to affect non-target organisms including humans, in the course of its application.
The effect of renal disease on plasma and urine depends on the proportion of glomeruli tubules affected and on the number of nephrons involved. Since endosulfan is a frequently used pesticide and there is incidence of toxic injury to the renal in relation to its widespread use.
The result of present study shows that exposure to endosulfan induced severe renal damages. The present findings suggested that endosulfan is a nephrotoxic chemical and that exposure produces significant renal toxicity, in rats.9
The histopathological studies also supported the protective properties of Amomumsubulatum. The areas of necrosis and ballooning degeneration of hepatocytes were observed in the toxic group. The group of animals pre-treated with Amomumsubulatum showed a marked protective effect with decreased necrotic zones and hepatocellular degeneration.10
As perAcharayaSushruthaandBhavaprakasha, BruhatEla is Vishagna, Bastivikarahara, Mutrakrichrahara, Mutraghatahara and also used in SarpaVisha.11
Hence,BruhatEla has taken to evaluate efficacy as it is having vishagna properties and it is also used in the bastivikaras.

7. Materials and Methods


    1. Source of data




  1. The literary source of present study will be collected from classical texts, modern texts,

published articles in reputed journals and from website.

  1. Raw Material BruhathEla.

  2. Albino Rats.



7.2 Methods of collection of data-
1. Raw drugs will be procured from the market and authenticated by Udupi Research Lab.

2.Endosulfan was purchased from registered pharmacies.

3. Wistar rats will be procured from the licensed source.

A. Inclusion criteria:

Either sex of Albino rats of 8-12 weeks, weighing 200-250gm.



B. Exclusion criteria

Albino rats less than 8-12 weeks, weighing <200-250gm.

D. Dosage-

Dose of animals will be calculated by following Paget and Barrens table. Blood samples will be

collected through Orbital plexus for assessment of parameter.

E. Assessment criteria –

1. Haemogram

2. Renal Function Test, Serum Urea, Serum Creatinine, Uric acid

3.Histopathologicalexamination of Kidney

4. Blood chemistry- SGOT, SGPT, ALP

5. Blood sugar,. Serum total Cholestrol, Serum triglyceride




F. Statistical methods-

The data generated during the study will be analyzed using one way ANOVA followed by

Dunnetmultiple’t’ test.

Yes, the studies will be conducted on experimental animals and also needed lab investigations mentioned above
