2. To confirm the taxonomic affinity of our strains, Primers ITS5 and ITS4 (White et al. 1990) were used to amplify the approximately 540bp ITS, followed by sequencing with the same primers. PCR was carried out on a PT-200 PCR thermal cycler (MJ Research, Waltham, MA) and products were purified with ExoSAP-IT reagent following the manufacturer’s instructions (USB Corporation, Cleveland, OH, USA). Bidirectional sequences were obtained using BigDye Terminator cycle sequencing kit (Applied Biosystems, Foster City, California). Products were analyzed on a 3100 DNA sequencer (Applied Biosystems). Sequences were edited and assembled using Sequencher 4.1 (Gene Codes, WI). ClustalX version 1.8 (Thompson et al. 1997) was used to align the sequences that were subsequently manually adjusted in MacClade 4 (Maddison & Maddison 2000). Italic accession number means sequences obtained for this publication. Further data to resolve isolates of Colletotrichum into three distinct groups were obtained from Rojas et al. unpublished results.
